How does Sildenafil work?
Sildenafil is a powerful selective inhibitor of cGMP-specific phosphodiesterase 5-th type (PDE-5). Realization of the physiological mechanism of erection is associated with the release of nitric oxide (NO) in the cavernous body during sexual stimulation. This, in turn, leads to increased levels of cGMP, the subsequent relaxation of smooth muscles of the cavernous body and increased blood flow.
Sildenafil has no direct relaxing effect on isolated human corpus cavernosum, but enhances the effect of nitric oxide (NO) by inhibiting PDE-5, which is responsible for the degradation of cGMP.
An essential prerequisite of the Sildenafil effectiveness is sexual stimulation.
When Sildenafil is prescribed?
Sildenafil is intended for treatment of erection disorders, characterized by inability to achieve or maintain penile erection sufficient for satisfactory sexual intercourse.
Before taking Sildenafil
You should not take sildenafil citrate if any of these conditions apply to you:
- hypersensitivity to sildenafil or any other component of the drug
- women and children aged 18 and less
Sildenafil should be used with caution in the following conditions:
- anatomical deformity of the penis (angulation, cavernous fibrosis or Peyronie's disease)
- diseases predisposing to the development of priapism (sickle-cell anemia, multiple myeloma, leukemia, thrombocytemia)
- diseases accompanied by bleeding
- acute peptic ulcer
- hereditary retinitis pigmentosa
- heart failure, unstable angina, myocardial infarction or stroke or life-threatening arrhythmia in the past 6 months, hypertension, hypotension
Application during pregnancy and breastfeeding:
Sildenafil is not intended for use in female patients.
How Sildenafil is taken?
You should take sildenafil citrate exactly as indicated by your physician. Do not exceed recommended dosages. Follow the directions of your prescription.
What should I do if I miss a dose?
Sildenafil is used as needed, so you are not likely to miss a dose. If you miss a dose of Sildenafil, take the missed dose as soon as you remember. If it is almost time for your next dose, skip the missed dose and take the medicine at the next regularly scheduled time. Do not take extra medicine to make up the missed dose.
What should I do if I overdose?
In single dose of the drug Sildenafil in a dose of 800 mg of adverse events were comparable with those in the drug at lower doses, but occurred more frequently. Treatment is symptomatic. Hemodialysis does not accelerate clearance of sildenafil, as the latter is actively bound to plasma proteins and is not displayed by the kidneys.
Which Sildenafil side effects are possible while the treatment?
As with any drug, there are some side effects while may occur while taking sildenafil citrate. However, not everyone who takes Sildenafil will experience side effects. In fact, most patients taking Sildenafil tolerate it well. During clinical trials there were reported the following sildenafil citrate side effects:
- nervous system: headache, dizziness, seizures
- cardiovascular system: flushing, palpitations, tachycardia, decreased blood pressure, fainting, nosebleed
- sensory organs: vision disorders, color vision, pain in the eyes, redness of eyes, ocular injection
- respiratory system: rhinitis
- digestive system: dyspepsia, vomiting
- skin reactions: hypersensitivity reactions, including rash

Which other medicines can interact with Sildenafil?
Make sure to inform your health care provide about prescription and over-the-counter medication, herbal supplements, and other medical preparations you are currently taking. Do not begin a new medication without letting your doctor know.
Sildenafil (sildenafil) may interact with the following groups of drugs:
- CYP3А4 and CYP2С9 inhibitors / inductors
- antiretroviral drugs
- nitrates, NO donors, nitrites
- alpha-adrenoblockers
- other ED drugs
How Sildenafil should be stored?
Store in a dry place at temperatures not above 30 C. Keep out of reach of children
